Quantitative Nano‐amperometric Measurement of Intravesicular Glutamate Content and its Sub‐Quantal Release by Living Neurons
نویسندگان
چکیده
Glutamate (Glu) is a crucial fundamental excitatory neurotransmitter released through vesicular exocytosis in the central nervous system. Hence, quantitative measurements of intravesicular Glu and transient exocytotic release contents directly from individual living neurons are highly desired for understanding mechanisms (full or sub-quantal release?) synaptic transmission plasticity. However, this could not be achieved so far due to lack adequate experimental strategies relying on selective sensitive nanosensors. Herein, we introduce novel electrochemical nanobiosensor based single SiC nanowire prone selectively measure real-time fluxes via by large vesicles ( ca. 125 nm diameter) present hippocampal axonal varicosities as well their content before exocytosis. Combination these two series revealed mode neurons, viz., only one third half molecules during events. Importantly, fraction remained practically same when were pretreated with L-Glu-precursor L-glutamine, while it significantly increased after zinc treatment, although both cases drastically affected. By affording inside its fractions work opening new possibilities investigating situ time regulatory glutamatergic neurotransmission

15 صفحه اولInhibition of synaptically evoked cortical acetylcholine release by intracortical glutamate: involvement of GABAergic neurons.
Cortical acetylcholine (ACh) has been shown to regulate diverse cognitive processes and its release can be regulated by neuromodulators that act presynaptically at cholinergic terminals. The neocortex receives dense glutamatergic input from thalamocortical and other fibres. The present study used in vivo microdialysis to examine, and pharmacologically characterize, the effect of glutamate on co...
متن کاملATP inhibits glutamate synaptic release by acting at P2Y receptors in pyramidal neurons of hippocampal slices.
It has been proposed that extracellular ATP inhibits synaptic release of glutamate from hippocampal CA1 synapses after its catabolism to adenosine. We investigated the possibility that at least part of this effect is mediated by ATP itself acting on P2Y receptors. ATP and various analogs decreased the amplitude and duration of glutamate-mediated excitatory postsynaptic potentials in all tested ...
متن کاملDetection of glutamate release from neurons by genetically encoded surface-displayed FRET nanosensors.
Glutamate is the predominant excitatory neurotransmitter in the mammalian brain. Once released, its rapid removal from the synaptic cleft is critical for preventing excitotoxicity and spillover to neighboring synapses. Despite consensus on the role of glutamate in normal and disease physiology, technical issues limit our understanding of its metabolism in intact cells. To monitor glutamate leve...
متن کاملSynaptic glutamate release by ventromedial hypothalamic neurons is part of the neurocircuitry that prevents hypoglycemia.
The importance of neuropeptides in the hypothalamus has been experimentally established. Due to difficulties in assessing function in vivo, the roles of the fast-acting neurotransmitters glutamate and GABA are largely unknown. Synaptic vesicular transporters (VGLUTs for glutamate and VGAT for GABA) are required for vesicular uptake and, consequently, synaptic release of neurotransmitters. Ventr...
